Antibody Engineering Services Cloning of variable domains ($250/clone, 1 week) Variable regions from the samples sent to us for sequencing will be cloned into a PCR cloning vector and confirmed by sequencing and restriction analysis. We deliver ~10μg of spin-column purified plasmid DNA and detailed project report containing the plasmid map, sequence trace files and annotated sequence of the insert. Synthetic antibody ($750/gene, 2-3 weeks) Client provides an accession numbers or sequences of the variable domains and the desirable antibody format. Genes will be recoded for optimal mammalian expression, synthesized using our block-based gene assembling technology, fused with desirable constant domains and cloned into our mammalian expression vector. We deliver ~10μg of spin-column purified plasmid DNA and detailed project report containing the plasmid map, sequence trace files and annotated sequence of the expression cassette. Single chain antibody ($500/gene, 2 weeks) Variable regions cloned by us or provided by Client will be reassembled into a single chain antibody format and cloned into our mammalian expression vector. We deliver ~10μg of spin-column purified plasmid DNA and detailed project report containing the plasmid map, sequence trace files and annotated sequence of the expression cassette. Chimeric antibody (from $1,000/construct, 2-3 weeks) Variable regions cloned by us or provided by Client will be fused with the requested constant regions and cloned into our mammalian expression vector. We deliver ~10μg of spin-column purified plasmid DNA and detailed project report containing the plasmid map, sequence trace files and annotated sequence of the expression cassette. Subcloning of antibody ($250/construct, 1-2 weeks) Any construct assembled by us can be subcloned into any vector provided by Client along with the signed liability exclusion form. We deliver ~10μg of spin-column purified plasmid DNA, description of the cloning strategy and annotated sequence of the insert. Custom designed antibody (please inquire) Any antibody can be synthesized or reassembled into any format and cloned into any vector desirable by the Client. Timeline and cost will be determined based on complexity of the project.
